Researchers at Helicos BioSciences have unveiled a new sequencing technology that can "read" individual DNA bases without the need for traditional amplification methods. This innovation significantly reduces the risk of errors during the sequencing process and makes whole-genome analysis more efficient and accurate. The technique, known as "synthesis sequencing," was tested using the M13 bacteriophage genome. The DNA was first broken into smaller fragments, and each fragment was tagged with a short DNA sequence to help anchor it in place. Then, a DNA polymerase enzyme was used along with fluorescently labeled nucleotides. As the DNA strand was synthesized, a camera captured the fluorescence of each newly added base, allowing researchers to determine the sequence in real time. This approach eliminates the complex and time-consuming amplification steps typically required in DNA sequencing. As a result, the process becomes faster and more cost-effective. According to Helicos BioSciences, their new instrument can sequence an entire human genome in approximately eight weeks at a cost of $72,000. The machine itself is valued at around $1.35 million. This advancement marks a significant step closer to the ambitious goal of sequencing a person's genome for just $1,000. Timothy Harris, the lead author of the study, believes this target will be achieved within the next five years.
