The innovative approach used by Helicos is known as "synthesis sequencing." As part of their testing, the researchers used the M13 bacteriophage as a model. The genome was fragmented into smaller pieces, and each fragment was tagged with a short DNA sequence that helped anchor it in place. Then, a DNA polymerase enzyme, along with fluorescently labeled nucleotides, was added to the reaction. As the DNA strand was built, a high-resolution camera captured images of each newly incorporated base, allowing for real-time detection of single bases through their fluorescence. This technique bypasses the time-consuming and error-prone process of PCR amplification, which has long been a bottleneck in genomic research. According to Helicos, the new instrument can sequence an entire human genome in about eight weeks at a cost of $72,000. The machine itself is priced at $1.35 million, but the potential for future cost reductions is promising. Timothy Harris, the lead author of the study, is optimistic that this target will be achieved within the next five years.
